制限酵素とは?

制限酵素は特定の認識部位またはその近くで二本鎖DNAを切断します。
制限酵素は、サブユニット構造、補因子の要件、および切断の特異性に基づいて、4つのタイプに分類されます。
特にタイプⅡの制限酵素は認識配列の部位でDNAを切断するため、実験に非常に役立ちます。
当社でお取り扱いしている制限酵素はすべてこのタイプⅡ由来のものになります。

制限酵素の種類

 

認識配列から離れたランダムな位置でDNAを切断します。これらの酵素は、機能するためにATPとS-アデノシル-L-メチオニンの両方を必要とします。 (※こちらのタイプは取り扱っておりません)

 

認識配列内またはその近くでDNAを切断します。これらの酵素はATPを必要とせず、メチラーゼから独立しています。
タイプIIの酵素は、毎日の実験室作業に最も有用なエンドヌクレアーゼです。
当社でお取り扱いしている制限酵素はすべてこのタイプⅡ由来のものになります。

 

認識配列から約20〜25塩基対離れた位置でDNAを切断します。
これらの酵素は機能するためにATPおよびS-アデノシル-L-メチオニンの両方を必要とします。 (※こちらのタイプは取り扱っておりません)

 

通常メチル化によって阻害されるタイプI〜タイプIIIとは対照的に、修飾された、典型的にはメチル化DNAのみを切断します。(※こちらのタイプは取り扱っておりません)
